Why Standard FBS Interferes With Exosome Isolation
Standard FBS contains a substantial concentration of bovine-origin extracellular vesicles. When exosomes are isolated from cell-conditioned media, whether by differential ultracentrifugation, size-exclusion chromatography, or precipitation kits, bovine vesicles present in the serum supplement co-purify with your target exosomes. Because they share the same physical characteristics, this contamination is difficult to remove at the isolation stage.
Particle counting by NTA or flow cytometry will overestimate yield. Proteomic and lipidomic profiles of the exosome fraction will include bovine proteins and lipids. Western blot data for canonical exosome markers will be obscured. If your cell culture media contains standard FBS, your exosome data contains bovine vesicle contamination.
